Long-term Changes of Fibroblast Growth Factor 23-Klotho Axis 12 Months after Unilateral Nephrectomy in Sprague Dawley Rats
Fibroblast growth factor 23 (FGF23) and Klotho have a major role in mineral metabolism. Since little is known about the long-term changes of mineral metabolism in living kidney donors, our aim was to show whether unilateral nephrectomy has a long-term impact on FGF23-Klotho axis in a rat model of living kidney donation. Klotho, FGFR1 and NaPi-2a co-transporter mRNA levels in rat kidney tissue were determined by real time PCR method and compared between unilaterally nephrectomised and naïve rats 12 months after unilateral nephrectomy. Immunohistochemical staining of kidney sections was done to show Klotho expression and distribution. Our results showed that there is a down-regulation of Klotho, FGFR1 and NaPi-2a gene in remnant kidneys of uninephrectomized rats compared to controls which could be explained by nephron loss and renal dysfunction.
