New targets for prevention of fibrosis in acute myocardial infarction
We identified BMP1 isoforms of the Bmp1 gene in the plasma of healthy volunteers and patients with acute myocardial infarction (AMI). In plasma of patients with AMI the BMP1-4 isoform has been identified while Bmp1-3 expression was upregulated in human heart sections from patients with AMI. We developed and administered specific BMP1-3 and BMP1-4 monoclonal antibodies to rats with AMI and demonstrated significantly lower heart enzymes (troponin t, CK-MB) in the plasma of treated rats. The histological analysis of the heart muscle showed that a combination of both BMP1-4 and BMP1- 3 antibody significantly decreased the size and the composition of the scar. The echocardiographic analyses showed that 21 and 45 days after the ligation the ejection fraction in rats treated with the combination of antibodies was 35% higher than in control rats, and the level of heart fibrosis was significantly reduced. Targeting specific BMP1 isoforms as mediators of heart fibrosis may offer a new therapeutic approach for reducing progression of AMI related scarring.
