SPE method for separation of lipid classes: comparative study
A solid phase extraction method (SPE) was developed and validated to enable the separation of lipid mixture into individual classes in high yield and purity. Three solid sorbents, based on styrene divinylbenzene/methacrylate copolymer (Nexus), octadecylsilica (C18) and aminopropylsilica (NH2) were compared as effective tools for the SPE separation of lipid classes: polar lipids (PL), sterols, monoacylglycerols (MAG), diacylglycerols (DAG) and triacylglycerols (TAG). The recoveries of lipids on Nexus were very low and this sorbent can not be used in future analysis of above-mentioned compounds. The obtained results suggest that aminopropylsilica SPE columns, used with appropriate solvent regime, are more effective as a sorbent and extracting tool for the separation of examined lipids into the classes and that the proposed method can be used as micro-procedure for a quick lipid preparation in analysis of edible oils, biological tissue samples and in regiospecific analysis of TAG by 1, 3-specific lipase hydrolysis.
