Food additives as alternative treatments for Nosema ceranae disease

Honeybee colonies affected by microsporidium Nosema ceranae is considered as an emergent disease that is posing a major threat to the health. Due to its detrimental effects on both strength and productivity of honeybee colonies, an appropriate control of this disease is advisable. Prolonged fumagillin treatment may contribute to developing resistance and declining concentrations of this preparation may exacerbate disease rather than suppress it. Furthermore, the use of antibiotic is not allowed in the EU member states. For that reason, the need for the utilization of food additives arises as alternatives to control this disease. In the present study three food additives with herbal, protein and mineral basis (NozevitPlus, BeeWell Aminoplus and Eko ZeoPet), as potential therapeutic agents, have been tested to control nosemosis type C in honey bee colonies. Also, the histopathological structure as well as content and distribution of mucosubstances and histochemical activity of aminopeptidase and non-specific esterase, both important enzymes of intermediate metabolism in the mid gut of honeybees, were investigated. The number of Nosema sp. spores was determined by counting spores using a Bürker-Türk haemocytom­eter, and to identify the species of Nosema sp. molecular diagnostic methods were used. For the purpose of describing neutral, acid and sulphate mucopolysaccharides we used the Periodic Acid-Schiff Reaction, Alcian blue-specific and Toluidine blue-specific staining. Degreased cuts of the midgut were stained according to the HE method in order to determine the general morphological characteristics of the tissue, and stained with special stains for determination of enzymes activity. In order to assess and verify the differences in the spore load data between experimental and control group at different sampling dates, one-way analysis of variance (ANOVA) and Mann-Whitney U test were performed using statistical software package Statistica-StatSoft v.7. Accumulating evidence from preliminary studies and carried out multiple administrations of different food additives on diseased honeybee colonies yielded a noticeable decrease in the number of N. ceranae spores in the midguts of honeybees originated from experimental group. Obtained results suggest oral natural zeolite clinoliptolites supplementation therapy is associated with significant therapeutic effects on nosemosis type C without altering normal bee physiology. However, further research is necessary to clarify the proposed mechanisms of action of the tested herbal, mineral and aminoacids compounds and to confirm the promising results observed in this study.
