Polar lipid fatty acid composition changes induced by lipid supplemented diets associated with mice liver regeneration

Regenerating liver is an ideal model in which to study the mechanisms that control cell proliferation and, in other words, provide better understanding of the mechanisms of neoplastic and some liver diseases. The purpose of this study was to investigate the influence of monounsaturated oil (MUFA-diet) such as virgin olive oil (VOO) and polyunsaturated oil (PUFA-diet) such as corn oil (CO), on the fatty acid composition of polar lipids in mice liver after partial hepatectomy. The relationships between changes in each fatty acid concentration, unsaturated/saturates (UFA/SFA) ratio, polyunsaturated/monounsaturated (PUFA/MUFA) ratio, PUFA/SFA ratio, C18:2n-6/C20:4-6 ratio and C22:6n-3/C20:4n-6 ratio in the different stages of liver regeneration (24 hrs, 48hrs, 7 days) were also determined. The fatty acid composition in the polar lipids was determined in 2-3 mo old Male Balb/c mice. Mice were separated into three groups (n=6) without surgical treatment and with different diet intake, and three diet depending groups (n=6) were 1/3 partial hepatectomized and sacrificed 24 hrs, 48 hrs and the 7th day after the operation. The groups were fed diets containing 5% (w/w) olive oil (FOO), 5% (w/w) corn oil (FCO) and standard pellet (control). Total lipids were extracted according to Folch et al., and polar lipids were separated and purified by solid-phase extraction and analysed in the form of fatty acid methyl esters by gas chromatographic (GC) analysis. Differences among groups were tested by nonparametric Kruskal-Wallis and Mann-Whitney U-test (p ƒŹ 0.05). Similar to total lipids, the fatty acid composition in the phospholipids was affected by fatty acids from different types of dietary oils in liver tissue. Palmitic (16:0) and stearic (18:0) fatty acids were major SFAs, linoleic (C18:2n-6), docosahexaenoic (C22:6n-3) and arachidonic (C20:4n-6) were major PUFAs, and oleic acid (C18:1n-9) was major MUFAs in polar lipids in the control and hepatectomized liver. The major changes in UFA/SFA ratio, PUFA/MUFA ratio, PUFA/SFA ratio, C18:2n-6/C20:4-6 ratio and C22:6n-3/C20:4n-6 ratio in the regenerating liver were found after 24 hrs under the partial hepatectomy. In the FOO and FCO groups after 24 hrs of liver regeneration, the UFA/SFA and PUFA/SFA ratios were increased and in the control group were decreased. In the FOO, FCO and control group after 24 hrs of liver regeneration, the C18:2n-6/C20:4n-6 and C22:6n-3/C20:4n-6 ratios were increased, and then decreased up to the 7th day. In the control groups during the liver regeneration, C16:0, C20:1n-9 and C20:4n-6 showed significant changes after 24 hrs, 48 hrs and the 7th day, C20:1n-9 showed a significant change in FOO groups, and C20:0 and C20:1n-9 in FCO groups. The smallest changes were found in FOO groups during the examined stages of liver regeneration. It is to conclude that the dietary enriched oils, type PUFA and MUFA affected the fatty acid composition in the total phospholipids of the liver tissue in control groups and after partial hepatectomy. The experiments can help in description of the nutritional modulation of DNA replication in the first phase of gene activation after partial hepatectomy.
