The effect of bilirubin on the membrane lateral domain structure of mouse neuroblastoma cells

We have recently shown that bilirubin induced an increase of the most ordered lateral domain population of the fibroblast cell membranes, which could be a simplified experimental model to describe the bilirubin effect on blood-barrier membranes. In order to evaluate the possible role of membrane structure alteration induced by bilirubin in the neuronal compartment of brain, mouse neuroblastoma cell line (Neuro 2a) have been used. The lateral domain structure was determined in the cell membranes. For this type of cells, we could also show that the action of bilirubin induced an increase of the membrane population of the most ordered domain type. The cells have been labeled by the methylester of palmitic acid at the fifth acyl carbon by the doxyl group. EPR spectra have been measured. The deconvolution of the spectra was performed by the superimposition of the calculated domain spectra, with the corresponding weight factors (w), describing the membrane occupation of the particular domains. The order and dynamics of the lipid molecules in the particular domain was evaluated by the best fit between calculated and experimental spectra. A significant increase of w for the most ordered domain, accompanied by a corresponding decrease of the less ordered domain membrane population, was detected at 5 mM and appeared strongly amplified at 100 mM bilirubin. We have shown that the membranes of mouse neuroblastoma cells undergo a drastic alteration of membrane strucutring by the action of elevated bilirubin concentration. Therefore, we could hypotesise that similar neuron membrane organizational changes may occur in the neurons of hyperbilirubinemic newborn infants.
