Synthetic strategies for the preparation of peptide based intercalators
Novel phenanthridine based fluorophores functionalized via peptide backbone are prepared using two strategies: classical or solution-phase methods and by solid-phase peptide synthesis. The advantage of solid-phase synthesis is easy preparation of small peptide libraries. In development of molecular probes, peptide intercalators have a prominent role while they combine unique features of both interacting species. In our case, phenanthridine moiety serves as an anchor to biological target but also as a good fluorescent reporter group for DNA, RNA and proteins. The length of the peptide linker combined with properties of different aromatic moieties attached offers additional interaction possibilities. The aim is to develop fluorescent probes with tunable properties that specifically interact with DNA, RNA and proteins which could lead to development of sensors and new drug candidates.
