Insights into catalytic mechanism of an enzyme
Investigation of the mechanism of enzyme action is maybe the most important aspect of protein research. There is a plethora of methods available to study interactions of an enzyme with its substrates and/or inhibitors. Among those, X-ray crystallography stands out by giving the precise 3D picture of enzyme's active site. In principle, the higher the power of X-rays, the higher the resolution of protein's 3D-structure and more details to be observed. Synchrotrons, as the most powerful X- ray sources, provide in many cases atomic resolution 3D-structures and enable detection of details that could otherwise be obscured. The superiority of synchrotron over in-house X- ray source in study of enzyme's active site will be demonstrated on several protein examples.
