Rational approach for designing an optimal in situ gelling ophthalmic system
Improving precorneal retention is one of the main approaches to optimize topical ophthalmic delivery. The in situ gelling formulations, which undergo phase transition form liquid to semisolid gel upon exposure to physiological environment, seem to be a promising tool (1). The aim of this study was to develop improved topical ophthalmic formulation based on poloxamers P407 and P188, as thermoresponsive polymers, and chitosan, as a promoter of mucoadhesiveness and gel strength. The main goal was to minimize the total polymer content while retaining the advantageous rheological properties related to the formulation during the storage, prior to the use and upon the application to the eye surface. The formulation was developed using D-optimal statistical design taking into account the critical formulation attributes, the temperature of gelation and viscosity.
