One-pot synthesis of cyanine-amino acids conjugates and their interactions with DNA/RNA
One-pot synthesis of cyanine-amino acid conjugate was performed in mild conditions proceeding without formation of toxic and odorous mercaptomethane and agreeable with terms of green chemistry. Despite mixture of three reactants in one-pot reaction only one (desired) product was formed. The aqueous solution of new dye revealed strong hypochomic and moderate bathochromic effect in UV/Vis spectrum upon titration with ds- DNA and ds-RNA, whereby latter gave the strongest changes. CD experiments yielded significant changes in CD spectra of polynucleotides (<300 nm) as well as appearance of strong induced CD spectra (ICD) at λ = 400-600 nm. At DNA/RNA over dye excess compounds bind as single molecules, while at higher ratios strong bisignate ICD typical for dye aggregate is observed, isoelliptic points supporting formation of one dominant type of complex formed. Intrinsically non-fluorescent, cyanine-amino acid conjugates upon titration with polynucleotides revealed emission strongly dependent on polynucleotide structure, AU-RNA yielding 7 times stronger intensity in respect to AT-DNA.
