Mutation in hemagglutinin-neuraminidase gene during attenuation of mumps virus
Adaptation of RNA virus wild type to unfamiliar host cell lines reduces their virulence and immunogenicity. Definition of the attenuation markers could help to select the master seed for vaccine strain and to develop in vitro test for monitoring of the mumps virus vaccine production. Mumps virus wild type was attenuated by serial passage in hens' eggs and human diploid cell culture. Analysed samples originated from the third passage on hens' eggs and from randomly selected passages on human diploid cell culture. SH and HN gene were analysed. Multiple sequence alignment of SH gene showed no attenuation markers. Multiple sequence alignment of HN gene showed existence of different quasispecies in different eggs from the 3rd passage on hens' eggs. Amnion 3p/1 was virus progenitor for samples on human diploid cell culture. They differed in five nucleotides. That can be used to develop in vitro test for monitoring of mumps virus vaccine production.
