Determination of the coding and noncoding nucleotide sequences of original Edmonston-Zagreb working seed
Edmonston-Zagreb vaccine strain is derived from a single clinical isolate of measles virus called the Edmonston strain and used for production of measles vaccine. Protein encoding and noncoding sequences of the original Edmonston-Zagreb working seed D242/99 were determined directly from DNA fragments generated by RT-PCR and sequenced by automated sequence apparatus. The sequence is compared to reference strains, i.e. Edmonston wild type AF266288 and Edmonston Zagreb AF266290. Edmonston-Zagreb working seed D242/99 and Edmonston wild type AF266288 differed in at least 30 nucleotide positions. Edmonston-Zagreb working seed D242/99 and Edmonston-Zagreb AF266290 differed in 7 nucleotide positions. We suppose that such substantial number of observed differences is due to additional four passage of Edmonston-Zagreb AF266290 in Vero cell culture (Parks et al., J. Virol. 75: 910-920, 2001)
