Application of monolithic chromatographic media in molecular detection of viruses
Monolithic media represent a novel generation of stationary phases used for liquid chromatography. The structure of monoliths provides better hydrodynamic properties compared to the conventional chromatographic supports. In this work the application of monoliths in molecular detection of viruses is described. Measles and mumps viruses were chosen as model viruses. Our results show that it is possible to bind viral particles and viral RNA on monolithic column. It is also shown that it is possible to concentrate viral particles from diluted suspensions (at viral dilutions 2 to 4 times below the detection limit of RT-PCR method). Presented results indicate possible applications of monolithic media in concentration of virus particles in order to improve the sensitivity of viral detection as well as for isolation of viral RNA from cell-free biological fluids.
