Seroprevalence of Leptospirosis in Different Species of Wild and Captive Reptiles in Croatia

Background: Leptospirosis is one of the most widespread zoonosis that can affect all groups of vertebrates. However, our knowledge of the role of reptiles in the epidemiology of leptospirosis is still scarce (Minette 1983, Oliveira et al. 2016). The aim of this study was to determine the prevalence of leptospiral antibodies in wild and captive reptiles and to use the obtained data to facilitate the study design for future research on this topic. Methods: Fifty-six blood samples of reptile species including 3 corn snakes (Pantherophis guttatus), 2 lizards (Pogona vitticeps and Iguana iguana), 34 slider turtles (Trachemys scripta) and 16 Loggerhead sea turtles (Caretta caretta) were tested by Microscopic Agglutination Test (MAT). MAT was performed using a panel of 12 Leptospira spp. serovars. Results: Antibodies against six different Leptospira spp. serovars were detected in 12 (21.8%) animals: corn snake, green iguana and 10 slider turtles. The most frequently detected reactions were against serovars Saxkoebing and Tarassovi at titres ranging from 1:50 to 1:400. Titre of 1:3200 against serovar Saxkoebing (followed by cross reaction to 9 other serovars in lower titres) was detected in a corn snake. A sample taken from the same animal a year later showed a positive reaction for the same serovar(s) in titres up to 1:100. Conclusions: This study confirms that investigated free living and captive reptiles of several species were exposed to Leptospira spp. Their exact role in epidemiology of infection as well as the dynamics of the infection in reptile species remains to be further investigated.
