Human decidual T lymphocytes express CD3-zeta

The T cell antigen receptor is associated at the cell surface with the CD3 complex of polypeptides (gamma, delta, eta and zeta). C 3-zeta is a transmembrane protein expressed in T cells and NK cells as a homo- or hetero-dimer. It is also bound in association with the transmembrane from FCgammaRIII (CD16), with CD2 and with p58 molecules in CD3- cells. The cytoplasmic domain of the CD3-zeta subunit is involved in signal transduction and subsequent activation of T cells. It has been shown that expression of CD3-zeta is very low or absent in patients with tumors and immunodeficiency. Our aim was to investigate CD3-zeta expression in human CD3+ and CD3- decidual leukocytes using a mAb that recognizes a cytoplasmic epitope of CD3-zeta. Fresh decidual tissue from elective first trimester termination of pregnancy (n=10). After isolation, cells wre immediately fixed in 4% paraformaldehyde. Before staining with different panels of mAb, cells were permeabilized with 0.1% saponin, and subsequent measurement of surface as well as intracellular antigens was performed by flow cytometry. We found that CD3-zeta was expressed in most decidual leukocytes. By simultaneous measurement of surface and intracellular antigens, approximately 70% of CD3+ cells was positive for CD3-zeta ; this was also the case for both CD4+ and CD8+ cells. A similar or even higher percentage of CD56+ cells was CD3-zeta+. Possible anergy of decidual T cells cannot therefore be ascribed to a deficiency of CD3-zeta, but could result from a lack of other signal transduction molecules. However, quantitive differences in CD3-zeta may influence decidual T cell function in pathological pregnancies.
