The effect of progesterone induced blocking factor (PIBF) in regulation of cytolitic potential of decidual lymphocytes

Decidual lymphocytes (DL) mostly express phenotype similar to NK cells (CD3-CD16-CD56bright+) with a high content of cytolytic molecule perforin in their granules. Previously we have shown that progesterone reduces the number of perforin + cells in the suspension of DL cocultured with decidual adherent cells (DAC) containing stromal cells and macrophages. Progesterone also decreases cytolytic activity of DL against K-562 targets. The aim of this study was to analyse whether these effects are progesterone specific and mediated by specific progesterone's mediator Progesterone Induced Blocking Factor (PIBF). DL isolated from human first trimester pregnancy decidua were cocultured with DAC in the presence of PIBF and the number of perforin positive cells was analysed by flow cytometry. Cytolytic activity of progesterone or PIBF treated DL were measured by 2hr PKH-26 (red) cytotoxicity test against NK sensitive K-562 cells. Anti-PIBF antibodies reverted progesterone mediated immunosuppression. PIBF+ cells were analysed by immunohistochemistry. PIBF reduces perforin expression in DL cocultured with DAC and in a dose dependent manner decreases cytolytic activity of DL. It blocks perforin egzocytosis from DL in the contact with K-562 targets. Anti-PIBF antibodies reverted progesterone mediated suppression of cytolytic activity. Significant number of PIBF+ cells were found in the first trimester pregnancy decidua. Our results clearly show that PIBF affects specifically perforin expression and cytolytic activity of DL and possibly indicate the importance of this mechanism at the materno-fetal interface.
