Involvement of Fas/Fas ligand mechanism in TH1 mediated cytolytic activity of peripheral blood lymphocytes of pregnant women
Successful pregnancy is characterized by the prevalence of Th2 cytokines over Th1 cytokines, which modulate systemic immune response of pregnant women in a way to downregulate cell mediated immunity. In spite of this pregnant women are not more susceptible to various infections compared to nonpregnant, and their peripheral blood lymphocytes (PBL) can increase cytotoxicity following Th1 cytokines stimulation. Lymphokine activated killing (LAK) of both T lymphocytes and NK cells can be mediated by different mechanisms as increased binding of effectors to targets or upregulation of cytolytic mediators expression. The aim of this study was to investigate the involvement of Fas/Fas ligand (FasL) mechanism in LAK activity of PBL of pregnant women stimulated by Th1 cytokines. Peripheral blood mononuclear cells (PBMC) of first trimester normal human pregnancy were separated by density gradient centrifugation and stimulated with IL-2 (1000 iu./ml), IL-15 (5 ng/ml), IL-18 (10 ng/ml) or IFNg (1000 iu./ml) for 3 days. Cytolytic activity of PBL, as nonadherent cell fraction of PBMCs, was analysed against NK resistant P815 targets and P815 Fas transfectant cell line by PKH-26 red cell linker kit and flow cytometry. IFNg and IL-18 were unable to induce LAK of PBL neither against P815, nor against P815 Fas transfectants. IL-2 and IL-15 induce LAK of PBL against P815 with the same efficiency. IL-2 as well as IL-15 stimulated PBL lyse significantly stronger P815 Fas transfectants than P815 targets. Th1 cytokines differ among themselves by capability of inducing LAK activity and by effector mechanisms used in these processes. IL-2 and IL-15, in concentrations cited, induce the same level of LAK of PBL of pregnant women against P815 cells, and both IL-2 and IL-15 stimulated PBL use FasL in killing P815 Fas transfected targets.
