The effects of short and long term TH1 cytokine stimulation on Fas/Fasl mediated cytotoxicity of first term pregnancy peripheral blood lymphocytes
The dominance of Th2 cytokine secretion (IL-4, IL-5, IL-6, IL-10) over Th1 cytokine secretion (IL-2, IL-12, IL-15, IL-18, TNF , IFN ) is found in peripheral blood lymphocytes (PBL) of pregnant women. It is considered that Th2 cytokines support successful outcome of pregnancy by attenuating cell mediated immunity. However, pregnant women are not more susceptible to various infections comparing to non-pregnant women and their PBL can increase cytotoxicity following Th1 cytokine stimulation. The aim of this study was to analyse the involvement of Fas/FasL mechanism in lymphokine (IL-2, IL-12, IL-15 and IL-18) activated killing (LAK) of PBL of pregnant women. Peripheral blood mononuclear cells (PBMC) of first trimester normal human pregnancy were separated by density gradient centrifugation and stimulated with IL-2 (1000 IU/mL), IL-12 (50pg/mL), IL-15 (5 ng/mL), (IL-18 10 ng/mL) for 18 and 72 hours. Cytolytic activity of PBL as nonadherent cell fraction of PBMCs was analyzed against NK resistant P815 target cells and P815Fas transfectant cell line by PKH-26 red cell linker kit and flow cytometry. IL-2, IL-12 and IL-18 upregulate cytolytic activity of PBL stimulated for 18 hours against P815Fas cell line, while these cytokines do not influence cytotoxicity of PBL against P815 cells. Long term IL-2 and IL-12 stimulated PBL show enhanced cytolytic activity against both target cell lines, while IL-18 long term stimulation of PBL is not able to change cytotoxicity of PBL. Interestingly, IL-15 strongly augments cytolytic activity of 18 hours and 72 hours stimulated PBL against P815 and P815Fas cell lines. More importantly, IL-15 stimulated PBL lyse significantly more P815Fas transfectants than P815 cells. Th1 cytokines are capable of inducing LAK activity of PBL by using different mechanisms. While IL-18 seems to be short term cytotoxicity stimulator of PBL against P815Fas cells using predominantly Fas/FasL mechanism, IL-2 and IL-12 act as both short and long term cytolytic stimulators against P815Fas transfectans, and are able to lyse P815 targets only following 72 hours stimulation. However IL-15 is equally efficient as short and long term stimulator of LAK activity of early pregnancy PBL. These results indicate the potent influence of Th1 cytokines on Fas/FasL mediated cytotoxicity of PBL of pregnant women.
