Gene mapping in Mischanthus sinensis Anderss. using new mapping strategy "offspring cross"

A new mapping strategy named 'offspring cross' has been developed to construct the first genetic linkage map of the species Miscanthus sinensis Anderss. (2n = 2x = 39). Basically, this mapping strategy consists of two sequential crosses. First, two individuals contrasting for the traits of interest are selected and crossed to generate a first offspring from wich two individuals are chosen and crossed, being this progeny the mapping population. Consequently, both parents are fullsibs. A total of 383 RAPD markers were analysed in the complete population. Out of these, 257 were mapped into 28 linkage groups. The total length is around 1, 074.5 cM with an average density of 4.2 cM per marker. Both parents contributed equally to the map. In this way, 62 markers were inhereted from F1.1 (P1), 63 from F1.7 (P7) and 132 were bi-parental markers (markers segregation in a ratio 3:1). Besides, the same contribution of mono-parental (markers segregation in a ratio 1:1) and bi-parental markers was obtained. This facilitates the development of a single map for both parents in contrast with the normal situation in this type of mapping populations where a map for each parent is developed. This map has been successfully in quantitative trati loci (QTL) mapping for traits of interest such as mineral content and agronomic traits. The offspring cross mapping strategy is proposed to obtain a higher effeciency in developing integrated maps including both parents.
