Alcohol Effects the Structure of Human High Density Lipoproteins
We have studied the impact of different alcohols on the structure of two human HDL subfractions, HDL2 and HDL3. To follow the modulations in the lipid-protein interface induced by different alcohols, the ESR spectroscopy was applied on HDLs labeled with two different spin labels, one for protein and the other for lipid moiety of the particle. Since the experimental spectra of the SL covalently bound to the apoprotein indicate the mobility of predominantly SL hydrophobic apoprotein environment, the increase of the SL mobility upon alcohol addition reflects the opening of the protein structure. The influence of alcohol on the lipid monolayer spin labeled HDL reveals the decrease of the ordering in the lipids with the increase of the alcohol incorporation in the lipoprotein particle.
