The CD44H expression on breast carcinoma cells and lymph node metastasis

The expression of CD44H molecule a standard from of the main cell surface hyaluronan receptor, was analysed on the sulface of breast carcinoma cells by immunohistochemistry and flow cytometry. In order to investigate prognostic parameters such as tumor size, lymph node metastasis, mitotic index and tumor grades. Immunohistochemical analysis was performed on the frozen sections of 70 invasive carcinomas, among which 60 cases were of ductal NOS type, 6 ductal special types and 4 lobular carcinomas. In 13 cases (18.6%) most of tumor cells (>90%) were positive for the CD44 whereas the rest of them were either heterogeneous (38.6%) or negative (42.8%) in CD44 staining. Statistical analysis did not reveal any relationship betweell the CD44 staining pattern and prognostic parameters. Flow cytometric analysis was performed on 40 primary carcinomas and 20 tumors from the lymph node metastasis. The results were expressed as a percentage of CD44 positive cells, mean fluorescence intensity of positive cells and mean fluorescence intensity of the whole tumor population. Again there was no significant relationship with prognostic parameters but we found higher CD44 expression in ductal NOS carcinomas compared to other types (p<0.05). The tumors from lymph node metastases had greater percentage of positive cells with also higher fluorescence intensity than primary tumors: these results were also significant (p<0.05) human biopsy specimens leads us to the conclusion that any IL-10 mediated migratory effect might not be functionally related to MMP-expression.
