Effect of maltose on cell growth and amylase production by Lactobacillus amylovorus DSM 20531T
Microbial amylases are among the most important industrial enzymes and growing spectrum of their applications goes far beyond textile, food, and paper industry. Bacterial strain Lactobacillus amylovorus DSM 20531T uses maltose, starch and raw starch as substrates and over short growth periods produces extracellular amylolytic enzyme. In this work effect of maltose on growth of L. amylovorus DSM 20531T and concordant production of extracellular amylase was observed. Cultivation of these strain on maltose (three cultivations per week) resulted in decrease of specific growth rate ( = 0.51 h-1, first week ; = 0.35 h-1, third week) while significant effect on amylase production was not detected. Crude enzyme was purified by ammonium sulfate precipitation, dyalisis and ion-exchange chromatography to obtain pure enzyme. Three different protein peaks with amylase activity were detected and further characterized.
