Genetic diversity of ‘Candidatus Phytoplasma solani’ strains associated with Bois noir disease in Croatian vineyards
Variability of genotypes involved in Bois noir (BN) pathosystem, disease induced by ‘Ca.P.solani’ (CPs), was assessed by a multilocus sequence typing (MLST) approach. Genotyping was performed on almost ninety selected CPs strains detected in the scope of an eight-year national survey encompassing grapevine samples, weeds and insects from all Croatian viticultural regions. The extensive tuf, secY, vmp1 and stamp based MLST analyses revealed the presence of at least 19 different CPs comprehensive genotypes. The prevalent genotype identified is affiliated to the main vector Hyalesthes obsoletus and widespread in central north-western part of Croatia. In addition to H. obsoleteus, CPs was also detected in samples from the genus Cixius that grouped within distinct stamp/comprehensive genotype. These results revealed considerable genetic variability and the wide geographical distribution of CPs in Croatia. Furthermore, considerable number of comprehensive genotypes found also indicated presence of several independent epidemiological cycles included in the BN pathosystem.
