Cyclosporine A differently affects the function of the two Main T cell subsets

Cyclosporine A (CysA) is a potent immunosuppressive agent, widely used for prevention of graft rejection in organ transplantation. It exerts immunosuppressive effect in vivo primarily due to its influence on the function of T cells. At which point exactly CysA affects T cell function is still unknown. In the present study we have investigated the effect of Cys A on the T cell proliferation and IL-2 production by CD4+ -enriched and CD8+ -enriched T cells, isolated from the spleen of normal mice. We found that the proliferation of CD8+ T cells was much more sensitive on the Cys A presence than CD4+ T cell proliferation. On the other hand, IL-2 production by CD4+ T cells was more affected by the Cys A presence than IL-2 production by CD8+ T cells. We suggest that there are at least two different models of Cys A action – one affecting IL-2 production and the other affecting T cell proliferation.
