Hantaviruses cause cytopathic effect in continuous human embryonic kidney 293 cell line
Hantaviruses (HTV) cause two important syndromes, hemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS). HTV persistently infect cultured cells and their natural rodent hosts. Since now, there was no evidence that HTV per se cause obvious cytopathic effect (CPE) in continuous cell cultures. We infected human embryonic kidney cell line (HEK) 293 with seven different hantaviruses: Hantaan (HTNV), Puumala (PUUV), Dobrava (DOBV), Seoul (SEOV), Sin Nombre (SNV), Andes (ANDV) and Laguna Negra (LNV). All tested HFRS and HPS hantaviruses induced CPE in HEK293 cells at different time points. The higher concentration of virus gave clear, strong CPE with obvious cell destruction. Using plaque reduction assay we showed that the virus titer rapidly decrease with the CPE appearance. There are at least two general pathways for cell death: necrosis and apoptosis. Using TUNEL assay, we showed that both HFRS and HPS viruses induced apoptosis in HEK293 cells. Recently it was described that FasL induced Fas/Apo1-mediated apoptosis in HEK 293 routinely used to generate E1-deleted adenoviral vectors. So, we looked whether the members of tumor necrosis factor (TNF) superfamily were responsible for apoptosis caused by HTV. Using RiboQuant MultiProbe protection system we did not confirmed it at the gene expression level. Our findings could be used for the design of the new tests in HTV detection and diagnosis, as well as further research of HTV immunopathogenesis. Important role it can also have in the quality control of biologicals produced from animal tissues and cells.
