The Effects of Amphotericin B and Fluconazole on Macrophage Functions
In the present study we investigated the effects of Amphotericin B and Fluconazol on the functions of peritoneal macrophages from BALB/c mice treated with these antimycotics. For each antimycotic the therapeutic dose used in clinical practice and twofold higher dose were administered intraperitoneally once a day for three consecutive days. The control group was treated with saline in the same way. The ability of peritoneal macrophages was determined by trypan blue exclusion. The adherence capacity of macrophages to a plastic surface was determined and the adherence index (AI) was calculated. The ingestion of heat killed Candida albicans was quantitated microscopically and expressed as phagocytic index (PI). There was statistically significant increase of AI of macrophages with the therapeutic dose of both antimycotics. However, higher dose of Amphotericin B decreased AI of macrophages. The therapeutic dose of Fluconazol significantlly increased PI of macrophages, same as higher dose of both antimycotics. Positive immunomodulry effects of antifungal drugs may be clinically relevant especially in compromised patients who are predisposed to opportunistic fungal infections and require long-term antimycotic therapy.
