Synthesis of rosmarinic acid in transgenic callus of Coleus blumei Benth.
Efficiency in the transformation of the three hybrids of Coleus blumei using wild and mutant strains of Agrobacterium tumefaciens and A. rhizogenes was evaluated. Successful transformation was dependent on specific bacterial strain-plant genotype interaction. Of the bacterial strains employed, the wild type B6S3 gave the maximum response. Sixteen transformed callus and one hairy root lines were established. They differed according to growth rate and rosmarinic acid (RA) production. RA contribution in dry weight varied from 1.6% (line Bz1) to 12.4% (line Ac1). The composition of the nutrient medium, addition of sucrose, phenylalanine, ammonium, nitrate and phosphate ions, and presence of the growth regulators, casein hydrolysate and Aspergilus sp. mycelium influenced growth and rosmarinic acid production in transformed tissue lines. The intensity of RA synthesis could not be related with agrobacterial strain or plant genotype used for the induction of transgenic lines.
