New cyclodextrins for supramolecular chemistry and biological interactions

Cyclodextrins (CDs) are macrocyclic oligomers of glucose. Their properties as molecular hosts, their multifunctionality, and their unusual adaptability as oligosaccharides to selective modification, make them unique mesomolecular subjects for supramolecular chemistry and chemical biology. Our work investigates the use of modified CDs in three main areas: Self Assembly It has previously been shown that CDs substituted with hydrophilic hydroxyethyl groups on the secondary-OH side and hydrophobic thioalkyl chains on the primary-OH side, form bilayer vesicles in water. Will reversing the positioning of these moieties on the CD result in similar vesicles, but with the hydrophobic cavity of the CD now orientated towards the bilayer membrane? The synthesis of catanionic CDs which self assemble to form supramolecular nanoscale structures is also of interest. Based on this, new catanionic CDs are being investigated for self assembly, and to control assembly. Cell Targeting ; The introduction of a saccharide onto an amphiphilic CD has been shown to be a means of targeting the vesicles to protein lectins by multiple interactions (cluster effect). The introduction of varying glycosyl groups enable targeting to a wider range of cell receptors for possible CD drug-delivery systems. Gene Delivery ; Forming a DNA complex with a suitable nonviral vector is one of the main directions that has been taken in the field of gene delivery, due to the probable greater safety and versatility of synthetic vectors. Cationic amphiphilic CDs have been shown to transfect DNA and are the first oligosaccharide vectors. The development of new polycationic CDs for this role is in progress.
