Early pregnancy decidual lymphocytes lyse NK resistant targets by perforin and Fas/FasL cytolytic mechanisms
The prominent feature of early pregnancy decidua is infiltration of CD3-CD16-CD56bright+ NK (dNK) cells followed by smaller number of macrophages and T cells. dNK are activated and filled with cytolytic mediators. Their activity is regulated by complex cytokine network. We analyzed cytotoxicity of decidual and peripheral blood lymphocytes (DL and PBL) against NK resistant targets under influence of Th1 cytokines. Decidual tissue and peripheral blood cells were obtained from elective termination of normal human pregnancies (6-10 weeks). Decidual tissue was enzymatically digested. Decidual and peripheral blood mononuclears were obtained by density gradient centrifugation. DL and PBL were collected following 18 or/and 72 hrs culture in the medium or in the presence of Th1 cytokines. Cytotoxicity assay, direct and indirect immunofluorescency were analyzed by flow cytometry. Non-stimulated PBL did not lyse NK resistant P815 and P815Fas targets while PBL stimulated with Th1 cytokines lysed more P815Fas than P815 cells. Non-stimulated DL lysed K562, but also P815 and P815Fas cells and this cytolytic effect could not be increased by Th1 cytokine stimulation. Non-stimulated and stimulated DL had higher cytotoxicity against P815Fas than P815 cells. 7% of CD56bright+ cells expressed FasL following IL-15 stimulation. DL cultured in the medium are activated and able to lyse NK resistant targets by perforin and FasL cytolytic mediators.
