On the mechanism of immunostimulatory activity of monomeric peptidoglycans
Mitogenic activity of immunostimulatory peptidoglycan monomer (PGM) was tested in vitro in short term cultures of mouse lymph node cells containing different quantities of PGM. Cultures stimulated with phytohaemagglutinin (PHA) or concavalin A (Con A) served as positive controls. As expected, PHA and Con A amplified the incorporation of radioactive thymidine into lymphocytes. PGM showed no mitogenic acitvity. Only the lymphocytes of the cultures containing 1 microgram of PGM were weakly activated. The presence of PGM suppressed even the stimulatory activity of PHA and Con A. In conclusion, PGM very probably is not a mitogen, or is only a weak one at strictly defined dose in relation to the lymphocyte number.
