Background and purpose: Potential cardioprotection could result from complex, and possibly opposite biological effects of ethanol and polyphenols, as shown through conflicting results of various studies. Our aim was to determine and compare the overall effects of red wine and ethanol moderate consumption on: (1) isoalated rat heart function in normal conditions and during the postichemic reperfusion ; (2) aortic vasoreactivity in normoxia and during hypoxia ; (3) liver lipid peroxides. Materials and Methods: 24 male Wistar rats, were divided into three groups, that were given during four weeks, as an only drinking fluid: 1. water, control (C) ; 2. red wine, diluted to 6% ethanol v/v (RW) ; or 3. ethanol, 6% v/v (E). Their hearts were isolated and retrogradely perfused in the Langendorff mode and after basic measurements, hearts were subjected to global normothermic ischemia followed by reperfusion. The thoracic aorta rings were mounted in tissue baths and subjected either to normoxia, where endothelium dependent and independent relaxation was measured ; or 50 min hypoxia, followed by norepinephrine (NE) and acetylcholine (Ach) challenge under hypoxic conditions. Results: After 40 min of reperfusion, isovolumetric left ventricular developed and diastolic pressure were highest in the RW group followed by the C and lowest in the E group. Vascular response to Ach was similar in all groups. Hypoxia induced equally impairment of vascular function in all groups. Liver lipid peroxides did not differ significantly among the groups. Conclusion: Moderate drinking of red wine and ethanol showed opposite effects on myocardial recovery after ischemia-reperfusion injury in the isolated rat heart.
