Cytotoxicity of CD1a+ first trimester pregnancy decidual cells towards autologuos CD3+ cells
Problem: Cytolytic potential of CD1a+ cells towards autologuos T cells of first trimester pregnancy decidua was investigated. Methods of study: Decidual mononuclear cells (DMC) were obtained by enzimatic digestion and gradient density centrifugation. Cytolytic mediators were analysed in CD1a+ cells using cell permeabilisation method. CD1a positive magnetic separation was performed from adherent DMC fraction whereas CD3+ cells were magnetically separated from CD56-CD83- DMC fraction using α -CD6 MicroBeads. Purified CD1a+ and T cells were co-cultured and short and long term cytolytic activity of CD1a+ cells towards CD3+ cells was tested using PKH-26 red assay. Results: Decidual CD1a+ cells highly express CD14 and CD56 molecules and are equipped with surface NK receptors as well as cytolytic mediators. CD1a+ cells show cytolytic activity against autologuos CD3+ cells. The percentage of CD3+ cells decreases after 3-day co-culture with CD1a+ cells. Conclusion: Decidual CD1a+ cells seem to be important for the regulation of T cells presence in early pregnancy decidua.
