Selenium status and glutathione peroxidase activity in arsenic exposed subjects in eastern Croatia
Blood selenium (BSe), creatinine-adjusted urinary Se (USe) and arsenic (UAs), as well as glutathione peroxidase activity (GPx), were determined in adult subjects from two Croatian areas with disparate levels of As in drinking water (Osijek: 37.9 mug/L and Andrijaševci: 612 mug/L). BSe was determined using GFAAS, USe and UAs were measured with HGAAS and GPx activities were analyzed using a commercial kit (Ransel, Randox Laboratories, UK). Creatinine was determined in all urine samples with automated chemistry analyser using a colorimetric Jaffe rate method. The median BSe levels in healthy adults were 95.1 mug/L (N= 25, range 66.0-117.2 mug/L) in Osijek and 74.3 mug/L (N= 38, range 48.6-113.7 mug/L) in Andrijaševci (p<0.01), respectively. The median GPx activity was also significantly lower in subjects from the high-As location (35.9 vs 45.8 U/gHb ; p<0.01). While USe and UAs were positively correlated in the control group (Spearman rank r=0.41 ; p=0.067), a nonsignificant negative correlation was observed in the high exposure subjects (r=-0.14 ; p=0.417). These preliminary findings are in agreement with previously observed effect of high As intake on increased biliary Se elimination which could contribute to As-induced oxidative stress.
