Enumeration of heterotrophs in river water with spread plate method: comparison of Yeast extract agar and R2A agar

Heterotrophic bacteria play an important role in the process of decomposition of organic matter in river ecosystems. The objective of this study was to compare two different media and two incubation temperatures (22 C and 35 C) for isolation and enumeration of heterotrophic bacteria from river water. A total number of 56 samples of surface water from Krka and Sava Rivers (Croatia) was analyzed in duplicate by the spread plate method using Yeast extract agar and R2A medium. R2A medium and incubation temperature of 22 C were found to be slightly more efficient for enumeration of heterotrophic bacteria in the river water. Preliminary assessment of microbiological quality of surface water indicated a more pronounced anthropogenic impact on Sava than Krka River, as a consequence of municipal sewage emissions into the river water.
