Bacterial evolution during prolonged stationary phase

The feature of Escherichia coli to grow in batch culture up to five years, only with the addition of water, has been termed Growth Advantage in Stationary Phase (GASP) phenotype. This phenomenon has been observed in Gram-negative and Gram-positive bacteria monocultures. GASP phenotype is due to selection of beneficial mutations in two global regulators of transcription of rpoS and/or lrp genes, as well as gene rearrangement involving IS5 insertion sequences. The aim of this investigation is to analyze mixed populations of Lactobacillus brevis L62 and E. coli or Salmonella enterica serovar Typhimurium during the prolonged period of carbon starvation for the evaluation of appearance of GASP mutants. According to previous analyses of the competition growth of E. coli, mutants can result in four GASP phenotypes ; however, we determined a new phenotype that we named moderate. This new class is characterized by GASP mutants of enterobacteria which are capable of outgrowing the younger lactobacillus culture and then coexisting together in the same ratio during 18 days. Studying cell traits in mixed bacterial cultures during prolonged stationary phase gives us a new insight into their interaction and evolution in order to control pathogenic bacteria.
