Vibrios are Gram-negative bacteria that are widespread, and some species may cause disease in aquatic animals and humans. The occurrences of Vibrio sp. were investigated in the three aquaculture areas along the East Adriatic coast. The aims of the present study were: to determine the presence of Vibrio spp. in sea water and fish samples ; to identify the isolated strains to species level and to investigate if they were correlated with pathogenicity in fish and human. In total 72 samples of the water were collected in two seasons during two years period, as well as 90 samples of the skin and gill swabs of the European seabass (Dicentrarchus labrax). Vibrio strains were isolated on TCBS medium, and molecular methods were used for final Vibrio identification. The results show the occurrence of Vibrio at aquaculture areas in the South, Middle and North Adriatic. Aquaculture areas did not differ significantly in the number of isolated Vibrio in the sea. Seasonal results of isolated Vibrio showed statistically higher number of Vibrio in spring than in the autumn. Differences between aquaculture areas in the number of Vibrio from the skin and gill were significant only in the number of Vibrio from the skin at North Adriatic in the spring 2008. Difference in the number of Vibrio from the skin in relation to gill was only significant for Middle Adriatic in the spring 2008. All isolates were phylogenetically analyzed by DNA sequence analysis of the gyrB gene and identified as Vibrio alginolyticus. On the basis of our results, we conclude that Vibrio strains isolated from the aquaculture areas in the Adriatic Sea, may significantly contribute to the outbreaks of disease in the fish and humans. A long-term research program of occurrences of V. alginolyticus is needed as preventive measure to protect fish and human health.
