Spatial patterns of genetic diversity in Fraxinus angustifolia populations across Europe – application to conservation prioritization

Genetic diversity patterns across tree species’ ranges have important implications for their effective conservation because they may affect the potential of populations to adapt to changing environment. Aim of our study was to identify current genetic diversity patterns in a wind-pollinated Mediterranean tree Fraxinus angustifolia across its European range and to test for a specific geographical gradient of genetic diversity. We sampled 1006 individuals from 38 natural populations and used nuclear microsatellites to identify genetic diversity within and among populations. We tested for geographical gradients in genetic diversity using linear regression and for isolation-by-distance (IBD) pattern using Mantel test. Finally, we created a genetic landscape surface for visualization of spatial patterns of genetic diversity/distances across the examined landscape. We found centres of highest genetic diversity in West and North Mediterranean populations compared to East and South Mediterranean populations which represent the most important reservoirs for conservation of genetic resources for F. angustifolia. Genetic diversity decreased significantly with longitude confirming a clear West–East gradient of genetic diversity. We also detected a significant IBD pattern across Europe. Our results suggest that conservation strategies should be adapted to opposing species borders.
