Molecular dynamics based studies on human DPP III revealed determinants for its broad substrate selectivity
Dipeptidylpeptidase III (DPP III) is a zinc-exopeptidase which hydrolyses a number of biologically active peptides (e.g., endomorphin1, endomorphin2 and enkephalin), and also some synthetics substrates, e.g., ArgArg2naphthylamide (RRNA), by cleaving dipeptides sequentially from their N termini. Because of its physiological as well pathophysiological roles (contribution in the protein catabolism, regulatory role in the pain-modulatory system, and enhanced level of DPP III was found in several carcinoma). In order to find determinants for its broad specificity for peptides of varying lengths and composition, we performed a series of MD simulations based on studies on human DPP III and its complexes with different substrates.
