Preliminary characterization of microbial water quality and bacterial population of fish in the hydropower plant reservoir in Croatia
The formation of reservoirs for hydropower plant could cause environmental changes. The changes can affect microbial water quality and the biota in the new environment adapted from the natural conditions in previous river. The preliminary study was carried out on the characterization of microbial water quality and bacterial population of fish in the hydropower plant reservoir Lešće in Croatia from spring to autumn in 2012 (May, July, September and November). Microbial indicators: total viable counts (HRN EN ISO 6222:1999), total coliforms and Escherichia coli (Colilert®), enterococci (Enterolert-E®), and Pseudomonas aeruginosa (Pseudalert®), as well as physico-chemical parameters were determined. The bacterial population of the fish was analyzed using MALDI TOF (Bruker) mass spectrometry. The comparison of the results of the microbial water analysis at the three sampling depths at the station of permanent control hydropower plant reservoir Lešće showed that values of almost all microbial indicators increase from the surface to the bottom of the reservoir. It was trend in the study from 2011 also. The oscillations of the analysed indicators were determined in 2012 in relation to 2011. In general, significant aberrations were not determined. The bacterial population of the fish was composed of five identified genera. The bacterial genera were predominantly Aeromonas (61.5%), Acinetobacter (23.1%), Enterococcus (7.7%), Ewingella (3.8%) and Citrobacter (3.8%). Regarding the high presence of Aeromonas in the fish bacterial population, these genera could be typical bacteria, representing a substantial part of the culturable indigenous microbial community from this aquatic environment. The presence of organisms particularly from the genera Enterococcus was attributed to the human activities in and around the hydropower plant reservoir Lešće.
