The influence of Puumala virus on the micro RNA expression in human epithelial kidney cells
This study investigates the expression or secretion of selected microRNAs (miRNAs) in vitro in HEK293 cells infected with Puumala virus (PUUV) or transiently transfected with PUUV N protein, and ex vivo in urine samples of patients with hemorrhagic fever with renal syndrome (HFRS) infected with PUUV, using real-time PCR. Limited deregulation of miR-21 and miR-27a expression was detected in HEK293, while miRNA abundance in supernatants was aberrant. Contrary to our hypothesis, transient expression of PUUV N did not yield significant changes in miRNA expression. In urinary samples of HFRS patients and patients with acute pyelonephritis, miR-21 and miR-27a were more abundant, while let-7e could be HFRS specific. In mild patients with HFRS only miR-21 and let-7e were deregulated, whereas in severe patients miR-24, miR-27a, and miR-127 were also altered. Detected miRNAs could present HFRS biomarkers correlating significantly with urea and creatinine levels, which represent hallmarks of HFRS progression and severity.
