DPC4 tumor suppressor gene in human sporadic colon cancer ; loss of heterozygosity
The most frequently affected regions in colorectal tumors are chrosomes 17p and 18q, where DPC4 gene exist. The purpose was to examine the prevalence of DPC4 loss of heterozygosity (LOH) in sporadic colon cancer. We examined 36 samples of human sporadic colon carcinoma of Croatian patients and corresponding normal tissue. Each sample was tested for LOH using threee polymorphic markers. Informativity was observed in 97% of analyzed samples. LOH at the DPC4 locus was detected in 51% of informative tumors. We concluded that inactivation of the DPC4 gene play a role in a multistep process of colon cancer.
