Introduction The removal, storage and transplantation of kidney alters the homeostasis of interior milieu of the organ. Phase of cold ischemia is important and potentially modifiable determinant of shortened graft survival and posttransplant morbidity. Although hypothermia maintains minimal metabolic tissue request, certain amount of reactive oxygen species (ROS) is generated. Ideal cold storage solutions should maintain the viabilty of the organ and be able to minimize reperfusion induced free radicals injury. We analyzed the potential of widely accepted preservation solution, University of Wisconsin (UW), to reduce kidney tissue injury and increase antioxidant capacity in prolonged cold storage. Materials and methods Male Wistar rats (3 months old) were sacrificed, both kidneys were harvested and placed in container with cold saline. Aorta was cannulated and kidneys were flushed either with saline or with UW solution. Organs were stored at 4ºC for 6, 12 or 24 hours. Lipid peroxidation (LPO) was measured via the thiobarbituric acid color reaction for malondialdehyde (MDA). Protein carbonyl content (PCC) was determined spectrophotometrically after labeling with 2, 4- dinitrophenylhydrazine (DNPH). Superoxide dismutase (SOD) and glutathione peroxidase (GSH- Px) activity were determined using comercialy available kit. Results In all analyzed groups of prolonged cold ischemia UW solution treated kidneys showed significantly lower level of LPO and PCC compared to saline. Activity of SOD and GSH-Px were also lower in UW treated group and positively correlated with levels of LPO and PCC. We observed time dependant increasement of LPO, PCC, SOD and GSH-Px in saline and UW treated kidneys. Conclusions Our study demonstrates that UW as preservation solution decreases tissue injury and has antioxidative superiority to saline. Time dependent increase of tissue deterioration when treated with UW emphasizes the need to optimize preservation solutions.
