Sažetak:
Neutrophils are dominant cell population in the lungs of mice infected with Legionella longbeachae. To assess the importance of granulocytes, presumably neutrophils in murine L. longbeachae infection in vivo an anti-Gr1 antibody (RB6-8C5) was used to cause transient neutropenia in mice. Groups of at least three CB57BL/6 mice were intratracheally challenged with 103 cfu of bacteria. Two doses of anti-Gr1 antibody (RB6-8C5) were administered before and after the infection: higher (250 ug/mouse) and lower (25 ug/mouse) dose. The flow cytometry analysis of CD11b and Gr1 cells population in peripheral blood of control and neutrophil depleted mice 24 hours post- treatment showed successful depletion of Gr- 1+CD11b+ cells (neutrophils). Moreover, there was an increase in CD11b+Gr1- cell population (possibly macrophages) in anti-Gr1 (lower dose) treated mice compared control. Anti-Gr1 antibody treated infected mice showed significantly increased susceptibility to infection. Infected mice treated with higher dose of mAb showed 100% lethality while lower dose treated mice had shown 75% lethality. It was also shown that anti-Gr1 treated infected mice had higher bacterial burdens in the lungs 72 hours and 7 days after infection. Interestingly, bacteria were not observed in the liver. Histopathologic findings had shown extensive bronchopneumonia and erosion of the epithelial mucosa 72 hours post-infection in higher dose anti-Gr1 Ab treated infected mice, while in lower dose treated infected mice the histopathologic changes were less severe. The results indicate that Gr1+cells play an important role in susceptibility to and clearance of Legionella longbeachae infection in mice. Further analysis should be performed to investigate the mechanism of their action in more detail.
